Summary, etc Plants are known for centuries for their medicinal value; that allowed the establishment of a large number of anticancer drugs with a very high efficiency. In Morocco, they play a very important role in traditional medicine, which is one of the major sources for new potentially bioactive molecules. <br/> The work presented in this thesis is a part of the national strategy, promoting Moroccan medicinal plants by developing ancestral knowledge and specific knowledge of plant species.<br/> Four medicinal plants were selected, on basis of the ethnobotanical study and according to their uses in traditional medicine. Sixteen plant extracts were screened on a panel of human cancer cell lines including cell types from solid and haematological cancers origins as well as non-transformed murine fibroblasts. Cell viability assays revealed that at least one extract from each plant was able to exert cytotoxic activity against the majority of cell lines tested, the IC50 values of the active extracts were in most cases ≤ 30 μg/ml.<br/> Next we focused on the anti-leukemic and apoptotic-inducing effects of Rm-HE, who exhibited a dramatic effect on Jurkat cells by promoting cell cycle arrest and partially JNK-dependent apoptosis characterized by an increase in Fas-L levels together with activation of caspases 8, 3, 7 and 9.<br/> A detailed study of Mv-DF mechanism of action was also carried out, its effects on the distribution of cell cycle and induction of apoptosis were detected by flow cytometric analysis for both (Jurkat and Jeko-1) lines, accompanied by a sharp increase in the activity of the effector caspases 3 and 7 and a significant inhibition of anti-apoptotic proteins (Bcl-2 and Mcl-1) as well as the PARP inactivation after 48 hours of treatment.<br/> Regarding Ormenis erioplepis, cell viability measurements, have allowed us to calculate the IC50 values for the most promising extracts, particularly Oe-DF and Oe-HE on the most sensitive lines. Cytometry analysis accompanied by p-H2A.X levels monitoring, showed that both the Dichloromethane Fraction and Hexanic Extract induce DNA double stranded breaks (DSBs) accompanied by cell cycle arrest in G1 (Jurkat, Jeko-1 and LN229) and G2/M (PC-3) phases which is agreed with the caspase activity observed.<br/> A qualitative study of the chemical composition and the major bioactive compounds of Rm-HE, Mv-DF Oe-DF and Oe-HE were performed by GC-MS.<br/> Finally, the data obtained in this study should honor the ancestral knowledge and provide a real scientific support for the use of these plants by herbalists and traditional healers, while providing an immediate starting point for many future studies.
